【摘要】:Thisguideprovidesguidelinesfortheselectionofanimalspecies,dosageandsamplingconditions,samplingandscoringmethods,statisticaldesign,andanalysisofgenotoxicityassaysinwhichtheendpointmeasuredisthefrequencyofmicronucleatederythrocytesinmammalianbonemarrow.1.1Thisguideprovidesrecommendedguidelinesforperformingthemammalianinvivobonemarrowmicronucleusassay.Underappropriatetestconditions,measurementofthefrequencyofnewlyformedmicronucleatederythrocytesinbonemarrowprovidesaconvenientindexofchromosomaldamageinnucleatederythrocyteprecursorcells.Therationalefortheoccurrenceofmicronucleiinconjunctionwithchromosomaldamagehasbeendescribedpreviously(1).Thisguidedescribesconditionsunderwhichthefrequencyofmicronucleatederythrocytesinmammalianbonemarrowisanappropriatemeasureofinvivochromosomaldamage,andprovidesguidelinesforthedesignandtechnicalexecutionofassaysemployingthisendpoint.1.2Thefollowingguidelinesformammalianbonemarrowerythrocytemicronucleusassayshavebeenpublishedbyorganizationsconcernedwiththeevaluationofgenotoxicitytestdata.Thesereferencesshouldbeconsultedforrecommendationsondetailsnotcoveredindepthbythisguideandforrequirementsofspecificorganizationsorgovernmentagencies(2-6).1.3Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
